چکیده

Microelectrodes were used to measure simultaneously the effects of indole-3-acetic acid (IAA) on membrane potential and cytosolic pH of corn coleoptile cells. IAA caused an initial depolarization followed by hyperpolarization, the latter displaying rhythmic oscillations. The extent of the changes in membrane potential was dependent on IAA concentration, and hyperpolarization, but not depolarization, could be detected with concentrations of IAA as low as 10 nM. Membrane hyperpolarization was preceded by a decrease in cytosolic pH. The decrease commenced approximately 5 min after adding IAA and continued for 15-20 min before reaching a new steady state approximately 0.1 pH unit lower than the original pH. The decrease in pH was readily detectable after treatment with 0.1 muM IAA. Fusicoccin and acetate, which, like IAA, induce elongation growth, caused a similar drop in cytosolic pH and subsequent membrane hyperpolarization, the decrease in pH commencing within seconds. The addition of fusicoccin to IAA-treated cells resulted in a further cytosolic acidification and membrane hyperpolarization. The two substances probably change cytosolic pH via different mechanisms. The results imply that one of the primary effects of auxins in coleoptiles is to lower cytosolic pH.

Effect of K+ and Ca2+ on the indole-3-acetic acid- and fusicoccin-induced growth and membrane potential in maize coleoptile cells

The role of potassium (K(+)) and calcium (Ca(2+)) in the regulation of plant growth and development is complex and needs a diverse range of physiological studies. Both elements are essential for satisfactory crop production. Here, the effects of K(+) and Ca(2+) ions on endogenous growth and growth in the presence of either indole-3-acetic acid (IAA) or fusicoccin (FC) were studied in maize (Zea...

Translocation of radiolabeled indole-3-acetic acid and indole-3-acetyl-myo-inositol from kernel to shoot of Zea mays L.

Either 5-[3H]indole-3-acetic acid (IAA) or 5-[3H]indole-3-acetyl-myo-inositol was applied to the endosperm of kernels of dark-grown Zea mays seedlings. The distribution of total radioactivity, radiolabeled indole-3-acetic acid, and radiolabeled ester conjugated indole-3-acetic acid, in the shoots was then determined. Differences were found in the distribution and chemical form of the radiolab...

1-Naphthyl Acetate-Dependent Medium Acidification by Zea mays L. Coleoptile Segments.

Zea mays L. cv INRA 5a coleoptile segments ecidify the incubation medium on the addition of 1-naphthyl acetate (1-NA). The buffering capacity of the bathing solution increases during 1-NA stimulated medium acidification. The solution bathing the 1-NA treated coleoptile segment was analyzed by high performance liquid chromatography. A considerable amount of acetic acid was detected in the bathin...

Photobiology of phytochrome-mediated growth responses in sections of stem tissue from etiolated oats and corn.

The far-red reversibility of the phytochrome-controlled stimulation of elongation of coleoptile sections by low fluence red light has been characterized in subapical coleoptile sections from dark-grown Avena sativa L., cv Lodi seedlings. The fluence dependence of the far-red reversal was the same whether or not the very low fluence response is also expressed. The capacity of far-red light to re...

Enhancement of CO(2) Uptake in Avena Coleoptiles by Fusicoccin.

When Avena coleoptile segments are immersed in a solution containing H(14)CO(3) (-), the appearance of label in the tissue is stimulated approximately 3-fold by fusicoccin application. This effect is rapid (1-2 minutes lag time), dependent upon respiratory energy, inhibited by carbonyl cyanide m-chlorophenylhydrazone, but not appreciably altered by cycloheximide treatment. A large percentage of...
